Phenotypic Drug Discovery with High Content Screening

چکیده

Drug discovery in its early history used to be conducted mainly in a target-agnostic fashion, focusing on qualitative readouts from a living system -cells, tissue, or organisms -and asking the question, ‘Did this entity affect the phenotype?’. With the rise of molecular biology in the early 1980s and an ever more powerful genomics toolkit from the 1990s, there was a marked shift away from phenotype-driven approaches towards screening techniques that looked to discover molecules affecting a defined disease target in an efficacious way. Initial assays were often performed using isolated, purified proteins rather than cells with the assumption that a target-based approach was the most streamlined path to finding the next marketable drug.

Phenotypic distance measures for image-based high-throughput screening

High-throughput image-based screening (also termed high-content screening) has become a popular method in systems biology, functional genomics and drug discovery. Data analysis of high-content screening can be divided into two steps: image quantification and phenotypic analysis. Previously we have developed two R packages, EBImage and imageHTS for image quantification. The current R package phe...

Bi-content micro-collagen chip provides contractility-based biomechanical readout for phenotypic drug screening with expanded and profiled targets.

Phenotypic screening has regained momentum in the pharmaceutical industry owing to its success over target-based screening. Most phenotypic screening relies on nonspecific biochemical readouts regarding cellular viability, which hampers the discovery of novel drug mechanisms of action (MOAs). Here we present a Contractility-based bi-Content micro-Collagen Chip (3CChip), which establishes cellul...

High Content Screening in Drug Discovery

The term high content screening (HCS) has become synonymous with imaging screen using automated microscopes and automated image analysis. The term was coined a little over 10 years ago, since then the technology evolved considerably and has established itself firmly in the drug discovery and development industry. Both the instruments and the software controlling the instruments and analyzing th...

Quantitative phenotypic and pathway profiling guides rational drug combination strategies

Advances in target-based drug discovery strategies have enabled drug discovery groups in academia and industry to become very effective at generating molecules that are potent and selective against single targets. However, it has become apparent from disappointing results in recent clinical trials that a major challenge to the development of successful targeted therapies for treating complex mu...

Drug Discovery for Neglected Diseases: Molecular Target-Based and Phenotypic Approaches Miniperspectives Series on Phenotypic Screening for Antiinfective Targets

Drug discovery for neglected tropical diseases is carried out using both target-based and phenotypic approaches. In this paper, target-based approaches are discussed, with a particular focus on human African trypanosomiasis. Target-based drug discovery can be successful, but careful selection of targets is required. There are still very few fully validated drug targets in neglected diseases, an...

Applications of High Content Screening in Life Science Research

Over the last decade, imaging as a detection mode for cell based assays has opened a new world of opportunities to measure "phenotypic endpoints" in both current and developing biological models. These "high content" methods combine multiple measurements of cell physiology, whether it comes from sub-cellular compartments, multicellular structures, or model organisms. The resulting multifaceted ...
